SUBSTANCE: invention relates to recombinant plasmid encoding hepatitis B virus surface antigen (HBsAg), it preparing and the strain of yeast Hansenula polymorpha as a producer of hepatitis B virus surface antigen. Invention provides the development of recombinant plasmid pHPS2 of size 7922 nucleotide pairs and consisting of the following members: dimer of artificial operon HbsAs/a/ayw comprising of promoter region of operon AOX1, gene HbaAs of hepatitis B virus and transcription termination site, yeast operon ura3 of gene encoding orotidine-5'- phosphate decarboxylase, operon region AOX1, bacterial operon of beta-lactamase and bacterial replication initiation site of type Co1 E1. Invention involves also the strain Hansenula polymorpha as a producer of hepatitis B virus surface antigen. Invention provides the enhanced expression of HBsAg.
