FIELD: molecular biology, biotechnology, genetic engineering, veterinary science. SUBSTANCE: invention relates to recombinant plasmid DNA p31NC constructed in vitro encoding polypeptide RNC where amino acid sequence Met-Arg-Gly-Ser-His-His-His-His-His-His-Ile-Pro is linked by its C-terminal end with sequence of nucleocapsid protein (NC-protein) of SRRS virus. Plasmid DNA consists of chemically controlling (IPTG) promoter/operator of phage T5, synthetic ribosome-binding site, phage lambda terminator of transcription, β-lactamase gene and Bam HI/Hind III-fragment encoding 125 amino acid NC-protein of SRRS virus. The construction provides effective biosynthesis of polypeptide. Prepared recombinant polypeptide shows antigenic properties of SRRS virus. Prepared plasmid construction provides IPTG-induced biosynthesis of recombinant antigen of SRRS virus in cells of bacterium E. coli. The use of invention allows to expand assortment of synthetic polypeptides for preparing highly sensitive and specific agents for diagnosis of SRRS for using in area where virus population caused this disease circulates.
