FIELD: biotechnology, protein genetic engineering, proteins, biochemistry.
SUBSTANCE: invention can be used for preparing correctly folded chimeric protein comprising the insulin precursor. The chimeric protein comprises a peptidyl fragment with length 20-92 amino acids that shows 60% of identity with the first 20 N-amino acid residues SEQ ID N:1 or SEQ ID N:2, and peptidyl fragment of the insulin precursor containing chain A and chain B of human insulin. C-end of the first peptidyl fragment is bound with N-end of peptidyl fragment of insulin precursor using residue Arg or Lys. Insulin is prepared by contacting the expressed chimeric protein with chaotropic agent or mercaptan. Invention provides preparing the insulin precursor in bioactive conformation and to screen a peptidyl fragment able to improve stacking the insulin precursor.
