FIELD: biotechnology, medicine, vaccines.
SUBSTANCE: invention relates to a method for preparing live vaccine for prophylaxis of influenza. Method involves infection of chicken embryos with the vaccine influenza strain, collection of virus-containing allantois fluid, addition of a stabilizing agent, sterilizing filtration, pouring into ampoules and lyophilization. Stabilizing agent comprises specimens of the following components, g: sucrose, 135; lactose, 90; glycine, 30; sodium glutamate, 15; tris-buffer, 5 and sodium chloride, 7.5, dissolved in 0.7 l of sterile distilled water and mixed with 10% gelatin solution in the ratio = 3:1. In the lyophilization process ampoules with vaccine are kept at temperature -35°C for 5 h being for the first 2 h ampoules are kept under atmosphere pressure (50 ± 10) mm of mercury column and then at temperature -16°C for 30 h. Then a temperature regulator is switched off for 6 h followed by additional drying at temperature +28°C for 15 h, the following filling ampoules with argon and sealing. Prepared vaccine comprises reduced amount of protein that decreases the possibility for arising allergic response reactions. Invention provides the improved technological effectiveness and economy of process for preparing vaccine.
