FIELD: medicine, ophthalmology.
SUBSTANCE: method involves isolation of neural retina macular region from freshly enucleated eyes of vertebral animals, washing out and holding retina tissue in aqueous-saline solution followed by centrifugation. Supernatant is collected, its components are separated by electrofocusing method in sucrose gradient density at pH 3.5-10, temperature 4-60C and voltage 2000-5000 V for 72-96 h. Fractions of acid proteins are collected, sucrose and ampholines are removed by dialysis, protein solution is dried and purified by electrophoresis on 5.5-15% polyacrylamide gel followed by elution of a low-molecular fraction with value Rf = 0.7-0.9 from gel with deionized water at 4-70C for 2-5 days, dialysis of the prepared fraction in dialysis sacs with deionized water with penetration limit of pores 2-8 kDa, drying the prepared regulatory peptide and its dissolving in physiological aqueous-saline solution. Invention provides enhancing percent yield of regulatory peptides possessing the biological activity in superlow doses and isolation of highly purified polypeptides of low molecular mass.
EFFECT: improved preparing method of medicinal agent.
