FIELD: medicine, clinical biochemistry.
SUBSTANCE: invention relates to optimization of determination of human blood neopterin by HPLC method. Method involves extraction with mixture chloroform and methanol in the ratio 1:1. After centrifugation upper aqueous-methanol layer is removed, evaporated and dry residue is dissolved in 0.02 M glycine buffer, pH 8.4. Analysis by HPLC method is carried out in a reversed-phase column 100 x 4 mm filled with sorbent Diaspher C18 with particles size 5 mcm at the rate 500 mcl/min in isocratic regimen and detection at exciting and emission wavelengths 350 nm and 430 nm, respectively. As a mobile phase a mixture of acetonitrile, isopropanol and 0.01-0.05 M citrate buffer (1:1:98), pH 3.5-4.5, is used that provides the maximal fluorescence of neopterin. Method shows relative simplicity and effectiveness of extraction (98%), rapid assay of analyte and high reproducibility.
EFFECT: improved method of determination of neopterin.
