SUBSTANCE: invention proposes a method for purification of double-stranded DNA (dsDNA) for a mixture of other components comprising the natural or artificially added sequence A-G-A-A-A-A-A-A-A-G-G-A or A-A-G-A-A-A-A-A-A-A-A-G-A-A. Method involves carrying out the following steps: (a) passing a solution containing indicated dsDNA through a support containing a covalently bound a single-stranded oligonucleotide of the sequence T-C-T-T-T-T-T-T-T-C-C-T and T-T-C-T-T-T-T-T-T-T-T-C-T-T that is completely complementary to the specific sequence represented with dsDNA to be isolated and forming with the latter the stable triplex structure; (b) eluting the end DNA by washing out the support at alkaline values of pH, and (c) isolation of dsDNA from eluate. Realization of the invention provide possibility for preparing the significant amounts of pharmaceutically pure dsDNA that can be used, for example, in genetic therapy or in aims for vaccination. Invention can be used in medical-biological industry.
