FIELD: medicine.
SUBSTANCE: invention offers method of ovary tissue preparation for transplantation for female patients with malignant neoplasms of reproductive system organs. Method implies ovary tissue intake, tissue treatment with media, separation of cortical layer from medullar part, cryopreservation by tissue freezing in cryoviols from +4°C up to -9°C with stepwise cooling, crystallization initiation at -9°C and performance of stepwise cooling of 0.3°C/min up to - 40°C and 10°C/min up to -140°C, immersion in the liquid nitrogen cryobank with the temperature of -196°C and storage with subsequent defrosting of tissue for transplantation. The method essence is using Ftushing media with heparin (Medicult) to treat the tissue and then Sperm Preparation Medium (Medicult). The tissue is transported in ice using Sperm Preparation Medium (Medicult). Then the cortical layer is cut into fragments on cold desk and dehydrated for 95 min. at the temperature of +4°C through placement of tissue fragments in the cryoviols filled with solution of 1.5 gram-mol/l propanediol and 0.1 gram-mol/l saccharose. The process is followed by further cryopreservation. After defrosting the tissue is placed in the Sperm Preparation Medium (Medicult) at ambient temperature. It is left in this medium till transplantation, but not more than 20 minutes. Suggested method ensures maintenance of the ovary tissue architectonics, survival of the follicles and stroma cellular components with subsequent recovery of the ovary endocrine function after transplantation.
