FIELD: microbiology.
SUBSTANCE: what is described is a method for genotyping Bordetella pertussis from clinical samples, differing by the fact that a biological material is sequentially taken from a patient’s oropharynx with two tampons and then placed into a physiological solution. This is followed by DNA extraction, nested PCR with described primers, evaluation of results by electrophoresis of PCR products in horizontal agarose gel and multilocus antigen sequence typing according to scheme MAST-2 for detecting allelic variants of ptxP-fim3-prn gene fragments coding a promoter ptxP region of pertussis toxin, fimbrial Fim3 protein and pertactin.
EFFECT: development of a method for genotyping Bordetella pertussis according to the MAST2 scheme directly from clinical samples without isolating a pure culture of the agent on the basis of nested-PCR.
