FIELD: medicine.
SUBSTANCE: invention relates to medicine, more exactly to biophysics, biomechanics and can be used to determine rigidity of muscular fibre costamere. Separation of single muscular fibre is performed, it is fixed to bottom of liquid cell of atomic force microscope, resonance fluctuation frequency is determined on air. Cantilever is brought to liquid cell bottom, calibrating force curve is obtained and used to determine calibrating coefficient a (in m/A). costamere location is determined by scanning fibre surface. System is transferred into contact mode and force curves are obtained on prominences corresponding to costamere, by which cantilever deflection (in A) , generalized depth of punching (in m) are found with further calculation of real depth of fibre punching (hs) and real applied to it force (Fs) by formulas hs=x-y·a, Fs =y·a·kc, where hs -real depth of punching (in m), x - generalized depth of punching (in m), y - measured deviation of cantilevel (in A), a - calibrating coefficient (in m/A), ), Fs - real applied to fibre force (in H), kc - coefficient of cantilever rigidity (in H/m).
EFFECT: method allows to increase accuracy of determining rigidity of muscular fibre costamere.
