FIELD: biotechnology.
SUBSTANCE: method of modification of peripheral blood monocytes comprises isolation of fraction of peripheral mononuclear cells (MNC) from venous blood which is carried out using a double gradient of Percoll density: 1.064 g/ml and 1.032 g/ml, respectively, at room temperature of +20°C. Then the isolated monocytes are placed in complete nutrient culture medium for adhesion to the plastic. Then, upon the expiry of 2 hours the complete nutrient culture medium is replaced with adding of 20 ng/ml IL4 and 20 ng/ml GM-CSF. At that the modification of monocytes is carried out by non-hydrolysed analogue of adenosine NEC A (5'-N-ethyl-carboxamide-adenosine) at a concentration of 100 mcM in incubation of cells in a CO2 incubator in a humid atmosphere 5% CO2 at +37°C for 72 hours.
EFFECT: invention enables to carry out the modification of peripheral blood cells in order to improve their reparative potential during autologous transplantation.
