SUBSTANCE: invention refers to molecular biology and concerns a test system for protein kinase GSK3β inhibitor screening. The characterised invention represents a genetic construct presented on Fig. 2, prepared of an expression vector pET32a, and comprising a gene of a catalytic domain of protein kinase GSK3β cloned by sites of restriction endonucleases EcoRI and HindIII, and a gene of a substrate of aminoglycoside phosphotransferase aphVIII from Streptomyces rimosus cloned by sites of restriction endonucleases XbaI, wherein the phosphorylation site AphVIII Ser-146 (AVAEGS146VDLED→ASAEGS146VDLSD) is cloned. The test system can be used in pharmaceutics and medical chemistry, particularly in development, creation and validation of new biological targets and test systems for the purpose of searching chemical leader compounds which can be potential candidates to drugs for treating human diseases.
EFFECT: test system enables pre-screening in situ of GSK3β inhibitors - low-molecular compounds of various chemical classes able to penetrate into Ecoli cells and gram-positive bacteria.
