FIELD: biotechnology.
SUBSTANCE: strain of Brucella abortus 19 is cultured in an L-shape in a nutrient medium prepared on the basis of meat-peptone hepatic glucose-glycerol agar (1.3-1.5% agar) with the addition of 10-15% normal horse serum and streptomycin at a dose of 2.5-5.0 U/ml at 37-38°C for 4-5 days. The resulting culture is rinsed with physiological solution and inactivated at 85-90°C for 60 minutes. The suspended matter is centrifuged at 3000-5000 rpm for 15-20 min, the supernatant is removed, and the precipitate is washed twice with physiological solution and centrifuged at 3000-5000 rpm for 15-20 minutes. The molten phenol is added to the bacterial mass in a ratio of 1:1. The phenol solution is adjusted by the physiological solution to 5%. The phenolic suspension is centrifuged at 5000-10000 rpm for 30-40 minutes. The supernatant is settled and used as antigen.
EFFECT: method can be used in combination of serological tests for brucellosis to reveal additional chronically sick animals.
