FIELD: medicine.

SUBSTANCE: inventions relate to field of cell biology, pharmacology and medicine. Described method of cultivating stem cells, which includes their spontaneous differentiation. Method includes preparation of pectin solution with concentration to 5% and etherification degree not higher than 50%; its application on base, formation of pectin matrix is carried out in presence of solution of cross-linking agent of ionic type. As solution of cross-linking agent of ionic type composition is used, which contains soluble calcium salt, sodium chloride, and buffer system based on components, which are non-toxic for cells. Introduction after application of pectin solution on base of additional operations for removal of pectin excess, washing base and its drying makes it possible to form matrix of different thickness from hydrogel layer to thin film. Different bases can be used in method as substrate. In other version of method implementation pectin solution with concentration to 5% and etherification degree not higher than 50%, prepared on isotonic solution is used. Introduction of cell culture is performed before matrix formation with further mixing; cross-linking agent of ionic type (CLA), prepared on isotonic solution, is then added to obtained mixture with further mixing. Said version of method implementation makes it possible to more largely approach process of cell cultivation to natural conditions, under which cells are in three-dimensional surrounding by extracellular matrix molecules.

EFFECT: claimed methods considerably extend versions of their application in cell technologies as they make it possible to obtain coatings of various thickness and structure on surfaces of different types.
