FIELD: chemistry.
SUBSTANCE: invention relates to biotechnology and specifically to a method of producing purified antibodies or antibody fragment, containing a Fc region or Fc fusion protein, using an affinity chromatography matrix to which the antibodies or antibody fragment, containing a Fc region or Fc fusion protein, are bound. The method includes clarifying a mixture containing an antibody or an antibody fragment containing a Fc region or Fc fusion protein. Said mixture is loaded onto the affinity chromatography matrix containing an immunoglobulin-binding protein. The affinity chromatography matrix is washed with a washing solution containing arginine in the concentration range of 0.05-0.85 M and a halogen salt in the concentration range of 0.1-2.0 M, where said washing solution has pH in the range of 8.0-9.5. Impurities are removed from the affinity chromatography matrix. The antibodies or antibody fragment, containing a Fc region or a Fc fusion protein are eluted from the affinity chromatography matrix at a low pH.
EFFECT: invention enables to obtain purified antibodies or an antibody fragment containing a Fc region or Fc fusion protein with high output.
