SUBSTANCE: group of inventions relates to biotechnology. The invention discloses a method for the extraction and a method for the purification of capsular polysaccharides from S.aureus type 5 or type 8 cells, as well as a composition for producing vaccines. The S.aureus type 5 or type 8 cells are treated with an organic acid in acidic conditions at a lysis step, where the cells are not subjected to autoclaving or treatment with lysostaphin. The acid treatment produces a capsular polysaccharide with an O-acetylation degree of 60-100%. A method of purifying an extracted capsular polysaccharide includes treatment with DNase/RNase, diafiltration, anion-exchange chromatography, gel filtration and concentration. The purified S.aureus type 5 or type 8 capsular polysaccharides are used to produce vaccines.
EFFECT: inventions enable obtaining capsular polysaccharides with low content peptidoglycan impurities (less than 5 wt %), nucleic acids (less than 1 wt %) and protein impurities (less than 5 wt %).
