METHOD FOR IDENTIFICATION OF ANTI-ALLOGENIC HLA-G ANTIBODIES Russian patent published in 2016 - IPC G01N33/533

Abstract RU 2585091 C1

FIELD: medicine.

SUBSTANCE: invention can be used for identification of antibodies to allogenic HLA-G in blood serum. For this purpose, test cross compatibility (cross match) of donor's mononuclear leukocytes with recipient's serum. B-lymphocyte subpopulation are analysed by mononuclear flow cytofluorometry on binding of monoclonal antibodies HLA-G, and degree of expression of HLA-G on test and reference donor's mononuclear cells. At that, significant are subpopulations of CD3-HLA-G+ and CD3+HLA-G+, from which coefficient of suppressing (CP) in test setting with respect to control by formula: C P HLA-G = ( H L A G O P H L A G C O N T ) H L A G C O N T × 100, where HLA-Gop is total relative number of subpopulations expressing HLA-G: (CD3-HLA-G++CD3+HLA-G+) in test setting, %; HLA-Gcont is total relative number of subpopulations expressing HLA-G: (CD3-HLA-G++CD3+HLA-G+) in control setting, %. Presence of sensitization to allogenic HLA-G is diagnosed at negative value CPHLA-G.

EFFECT: using given method enables detecting allogeneic sensitisation to HLA-G with help of flow cytometry, determining subpopulation CD3-HLA-G+ and CD3+HLA-G+.
