FIELD: biochemistry.
SUBSTANCE: present invention relates to ligands for affinity chromatography based on different domains of protein A (SpA) Staphylococcus. Ligand contains either several domains (C), or several domains (B), or several domains Z of protein SpA. Each of said domains contains deletion 3 or 4 sequential amino acids from N-end, starting from position 1, or deletion 3 sequential amino acids from N-end, starting from position 2. Said domains of protein SpA in ligands can additionally contain amino acid mutation in position 29 to reduce Fab binding. Present invention discloses an affinity chromatography matrix containing one of said ligands attached to solid substrate. Invention also discloses a method for affine purification using said matrix. Ligand affinity HPLC, containing at least five domains with protein SpA, each having a mutation to reduce Fab-binding, as well as deletion 4 amino acids from N-end, starting from position 1, is stable in basic conditions.
EFFECT: present invention allows to reduce fragmentation when using these ligands and matrices in purification methods.
