FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology and can be used for producing antibodies. Method involves culturing a host cell containing nucleic acids which encode an antibody under conditions suitable for antibody expression, and recovering the antibody from a cell or culture medium. Antibody contains the first polypeptide and the second polypeptide, each of which contains in the direction from the N-end to the C-end, at least part of the hinge region of the immunoglobulin of the human IgG1 or IgG4 subclass, which contains one or more residues of cysteine, CH2- and CH3-domain of immunoglobulin. First and second polypeptides contain a mutation Y436A and additionally independently include i) mutations 1253A and H310A, or ii) H310A and H435A, or iii) H310A and H333A (numbering according to Kabat).
EFFECT: invention enables to obtain antibodies with reduced ability to bind FcRn, but does not affect binding to staphylococcus protein A.
