FIELD: chemistry.
SUBSTANCE: invention relates to a method of determining monosaccharides in vaginal fluid
and comprises using a gas chromatography method with mass spectrometry detection. Samples are prepared by drying 10 cm3 vaginal washing at 30 °C for 24 hours. A weighed portion of dry residue weighing 35 mg is treated with 300 mcl of a mixture of N, O-bis-(trimethylsilyl)-acetamide, trimethylchlorosilane, N-trimethylsilyl imidazole at a ratio of 3:2: 3 and 40 mcl of N, O-bis-(trimethylsilyl)-trifluoroacetamide, prepared mixture is held in a vial at 80 °C for 15 minutes. Obtained derivatives are examined on a chromatography-mass-spectrometer with a mass-selective detector at temperature of analytical interface 280 °C and evaporator temperature 280 °C. Separation is performed on a quartz capillary column HP-5MS. Further, method comprises heating at a rate of 10 °C/min to 130 °C, holding system - 0 min, then at a rate of 3 °C/min to 210 °C - 0 min, then at a rate of 15 °C/min to 240 °C - 0 min, at a rate of 3 °C/min to 265 °C - 10 min and at a rate of 15 °C/min to 295 °C - 15 min, analysis time - 70 Min, flow rate of carrier gas (helium) - 1 cm3/min, average linear speed of carrier gas - 37.6 cm/s, scanning range m/z 45.0-350.0 amu, volume of injected sample - 5 mcl. For identification of TMC-derivatives of monosaccharides from their mass spectra, database of NIST is used, which are part of standard data processing program of mass spectrometer.
EFFECT: use of method provides high-accuracy of determination of monosaccharides in vaginal fluid.
