FIELD: biotechnology.

SUBSTANCE: methods are described for detecting of nucleic acid target sequence in analysis with PCE-NH (hybridization absence depending on cleavage and elongation of the probing and labeling oligonucleotide (PTO)). The present invention uses hybridization inhibition by formation of a target-dependent elongated duplex. Thus, according to the present invention, detection can be carried out of target sequences even when the hybridized oligonucleotide (HO) is not split. Therefore, we can easily construct sequence of 5'-tagged terminal PTO portion, sequences of capturing and matrix oligonucleotide (STO) and NO and easy determine the conditions of the reaction.

EFFECT: detection of hybrid between STO and NO can be carried out in another vessel, not in the vessel to extend the STO.
