FIELD: biotechnology.
SUBSTANCE: group of inventions relates to detection of a target nucleic acid sequence by a POCH (PO cleavage and hybridisation) assay on a solid substrate. Invention detects target nucleic acid sequence by use of POCH assay, in which PO (probing oligonucleotide) hybridised with target nucleic acid sequence is cleaved and cleavage of PO is detected by hybridisation with CO (capturing oligonucleotide). Uncleaved PO is hybridised with CO immobilised onto solid substrate. Design of PO and CO is convenient and optimisation of reaction conditions is routinely easy. Where detection of signal on solid substrate is continuously performed along with repetition of cleavage of POs, number of POs cleaved is increased upon repetition number of cleavage reaction and signal is changed in parallel with number of POs cleaved. In this case target nucleic acid sequence can be detected in a real-time manner. In contrast, change of signal is not observed in absence of target nucleic acid sequence.
EFFECT: use of group of inventions allows simultaneous detection of multiple target nucleic acid sequences using a mark, even of one type.
