SUBSTANCE: invention relates to determination of nucleic acid sequences. Disclosed is a group of inventions, including a device and method for optical monitoring of nucleic acid sequencing, computer-readable medium with a computer program and program element, used in said method, as well as use of 5-methyl-(2-(2-nitrophenyl)propyl)-carbonate dUTP, 5-methyl-(2-oxo-1,2-diphenylethyl)carbonate-dUTP as blocker in DNA sequencing in said method. Device includes a substrate in form of wire gauze for binding molecules on surface; optical circuit, configured to direct exciting radiation onto a substrate, receiving, detecting radiation and directing splitting radiation onto substrate; solution with nucleotides and enzyme, where nucleotides contain a blocker. Method involves providing a substrate with a molecule, irradiation of substrate with exciting radiation, limiting exciting radiation using substrate, receiving and detecting fluorescence of excited fluorescent nucleotide label, irradiation of substrate with splitting radiation, limiting splitting radiation and providing solution with multiple nucleotides and enzyme.
EFFECT: invention provides improved method for determination of nucleic acid sequence.
