SUBSTANCE: group of inventions relates to a method and apparatus for sequencing by synthesis (SBS) using simultaneous visualization of DNA clusters using two or more colour channels. Method includes obtaining a sample containing a first and a second nucleotide; bringing the sample into contact with a first fluorescent dye bound to a first nucleotide complementary to the first nucleotide, and a second fluorescent dye bound to a second nucleotide complementary to the second nucleotide. First and second fluorescent dyes emit, respectively, a first emitted light in a first wavelength range in response to the first excitation light and a second emitted light in a second wavelength range in response to the second excitation light. First average wavelength determined for the first emission spectrum of the first fluorescent dye and the second average wavelength determined for the second emission spectrum of the second fluorescent dye have at least a predetermined separation between 50 nm and 100 nm. Multiplexed fluorescent light is collected simultaneously and the first nucleotide is determined based on the first wavelength band of the first color channel and a second nucleotide based on the second wavelength band of the second color channel. Sequencing device comprises a flow cell containing a sample with a first and a second nucleotide; lighting system and light collection system.

EFFECT: group of inventions provides higher sequencing capacity, reduced imaging time.
