FIELD: biotechnology.

SUBSTANCE: invention is a method for assessment of the viability of cellular material derived from the human umbilical cord and placenta, according to which cells in complete growth medium are immersed in a 96-well plate, 5000 cells into the first 8 wells, 2500 cells into the second 8 wells, and 1250 cells into the third 8 wells, 6 hours later, the medium is removed from the wells and 100 μl of culture medium without fetal calf serum are introduced into each well with 10 μl of 3-(4,5-dimethylthiazol-2-yl)-2,5-tetrazolium (MTT) solution 10 mg/ml in PBS (sodium phosphate buffer, represented by an aqueous solution of salts containing sodium chloride, sodium hydrogenphosphate, potassium chloride and potassium dihydrogen phosphate, in which the osmolarity and ion concentrations in the solution correspond to the concentrations in the human body - the isotonic buffer solution), then incubated for 1 hour at 37°C in an atmosphere of 5% CO2, after incubation, the medium is removed from the culture plate wells without cell capture, and 100 μl of dimethyl sulfoxide is introduced to extract intracellular MTT formazan, and the optical density is measured spectrophotometrically at a wavelength of 540 nm against dimethylsulfoxide where the viable cell material is referred to as III-VI passages with a signal of 6.5*105 opt.u. and 5 ths/cell higher.
