FIELD: medicine.

SUBSTANCE: invention relates to a method for morphofunctional analysis of platelets contained in platelet-rich plasma (PRP) or platelet concentrate (PC). The essence of the method is determination of platelets concentration (CTP, ths/μl) in PRP or PC. The intravital staining of platelets is carried out by a dye prepared by dilution of 5-15 mg of trypaflavin and 15-25 mg of acridine orange at a room temperature in 100 ml of phosphate buffer at pH of 7.2-7.4 by introducing a dye into PRP or PC at a rate of 200 μl of dye per 1 ml of platelet concentrate or 100 μl of dye per 1 ml of platelet rich plasma. After that, a study of the preparation with stained platelets is performed using a fluorescence microscope followed by determination of the average luminescence intensity (LIexperiment) of the first field of view of the microscope, in addition, the theoretical luminous intensity (LItheory) is determined according to the calibration curve or by the formula, reflecting the average luminescence intensity of the first field of view of the microscope with vitally stained cells of the sample with a given platelet concentration (CTP), provided that all cells of this sample (100%) contain granules. Further, the relative content of platelets with granules (Dp.g.) is determined by the formula Dp.g.(%)=LIexperiment/LItheory×100%, while the platelets suitable for clinical use are those having the value of Dp.g. of 35 to 75%.

EFFECT: application of the method allows to analyze the population of vitally stained cells with high accuracy.
