FIELD: medicine.

SUBSTANCE: invention refers to medicine, namely to regenerative medicine, and can be used for preparation of thrombocyte lysate with high content of growth factors. Venous blood of the patient is taken, blood platelet counts with granules are evaluated, and samples containing at least 35 % of thrombocytes with granules are taken. That is followed by two-step centrifugation of the blood sample, first at 300–500 g for 5–10 minutes to obtain a plasma fraction with thrombocytes without leucocyte and erythrocyte impurities, then at 700–1000 g for 5–20 minutes for deposition of thrombocytes. Supernatant is removed with subsequent resuspension of thrombocytes with a phosphate-saline solution of PBS with pH equal to 7.3, or isotonic 0.9 % sodium chloride until the platelet conglomerates disappear. Then thrombocytes are cryodestructed with subsequent centrifugation of thrombosed samples of thrombocyte lysates at 3000–4000 g for 10–20 minutes at temperature of 4–6 °C for removing all cell fragments from the final preparation.

EFFECT: method enables to obtain a thrombocyte lysate in a plasma-free plasma medium with a high content of growth factors by preparing a platelet concentrate in a certain mode in a plasma-free plasma without cell quality disorders (thrombocyte and granule level, morphofunctional thrombocyte activity, thrombocyte adhesive activity) with high content of growth factors (PDGF, EGF, FGF) and low pro-inflammatory factors.
