FIELD: biotechnology.
SUBSTANCE: immobilized biocatalyst is proposed for fumaric acid production. The biocatalyst is the cells of the mycelial fungus of the Rhizopus genus introduced initially as a spore into the polyvinyl alcohol cryogel and sprouted to form mycelium. At that, the spores are included into the cryogel in the process of its formation. Germination of spores was carried out in a medium containing glucose 100 g/l, yeast extract 5 g/l and tryptone 10 g/l, or margarine 10 g/l and corn stalk extract 5 g/l, or potato starch 50 g/l and yeast extract 10 g/l, together with salts - (NH4)2SO4 2.36 g/l, MgSO4×7H2O 0.2 g/l, ZnSO4×7H2O 0.07 g/l, K2HPO4×3H2O 1.0 g/l, or in a medium containing barley malt 20 g/l or sucrose 30 g/l, yeast extract 10 g/l and soybean flour 10 g/l. The immobilized biocatalyst formed as a result of spores germination has the following composition (wt %): mycelium biomass (dry weight) - 6.5÷16.3; polyvinyl alcohol - 2.1÷6.0; water - up to 100.
EFFECT: biocatalyst is stable in acid medium, characterized by high productivity, provides an increased yield of the target product, the period of use in the batch process reaches 2100 h.
