FIELD: biotechnology.
SUBSTANCE: strain of bacteria Escherichia coli BL21 DE3 pClcRFP is suggested for detection of copper ions, biosensor and method for detecting copper ions in the liquid medium under analysis. The biosensor includes a housing with microchannels for the liquid medium under analysis, inside of trap microchannels in form of a series of micropillars with immobilized agarous spheres. Moreover, the agarose spheres contain genetically modified Escherichia coli BL21 DE3 pClcRFP bacterial cells, which are capable of synthesizing the fluorescent protein RFP. The method is carried out by means of the above biosensor and includes immobilizing bacterial Escherichia coli BL21 DE3 pClcRFP cells into agarous spheres with diameter of 50 to 70 mcm, placing them in biosensor traps, feeding the analyzed liquid medium into biosensor, measuring the glow of the agarous spheres, processing and comparing the results with the control ones.
EFFECT: inventions provide detection of copper ions in medium and an assessment of contamination degree using a small sample volume and a small amount of genetically modified bacterial cells necessary for the detection of one sample.
