FIELD: biotechnology.
SUBSTANCE: set is designed to identify the DNA of the Blastomyces dermatitidis irritant by polymerase chain reaction with hybridization-fluorescent results. The present set contains a BDags-2P probe constructed as a molecular beacon, and BDags-1F and BDags-2R oligonucleotides complementary to the α-1.3-glucan synthase gene fragments B. dermatitidis and having direct and reverse primer activity in the amplification reaction. The present invention is characterized in that the noted oligonucleotides have the following structure: 5'-GCG TTA TTGAAG CCG ATC CT-3' and 5'-AAC AGC CCG ATT GTC CAA AG-3', respectively. The present probe provides fluorescent detection and has the structure of 5'(FAM)-GGGATGCAGCGCAATTGCAATCCC-(BHQ1)3', where FAM is the carboxyfluorescein fluorescent dye, BHQ1 is the fluorescence quenching agent.
EFFECT: invention makes it possible to identify the DNA of the B dermatitidis irritant with high sensitivity and specificity.
