FIELD: biotechnology.

SUBSTANCE: polynucleotide library for production of coupled antibody sequences, a method for production of a polynucleotide of interest, and a method for analysis and application of sequencing data are provided. The polynucleotide library consists of compositions containing cDNA encoding heavy and light chains of the immunoglobulin variable region of the same clonal family. At that, each composition is in a separate container and a plurality of compositions are in a plurality of separate containers. Each composition thus comprises: (i) cDNA molecules derived from a single sample containing cDNA molecules encoding the immunoglobulin heavy chain variable regions and cDNA molecules encoding the immunoglobulin light chain variable regions from one or more B cells; and (ii) the identification portion of the sample attached to cDNA molecules. At that, the nucleotide sequence of the identification portion of the sample differs from the nucleotide sequence of the identification portion of the sample of other compositions present in other individual containers in the library.

EFFECT: presented inventions can be used to produce coupled antibody sequences and screen the molecules of interest.
