FIELD: agriculture.
SUBSTANCE: invention relates to agricultural biotechnology. Invention is a method of producing transgenic wheat plants by via a bioballistics method, including cold shock (low-temperature treatment of donor plants), cultivation of explants before transformation on a medium for inducing callus formation, wherein the medium contains picloram, osmotic treatment of the explants before transformation, wherein the explants are placed on a medium with a high osmotic pressure, in order to cause plasmolysis, introduction into cells of explants of foreign DNA using a PIG (particle inflow gun), proliferation of transformed cells without selective sampling, regeneration and selective sampling of transgenic shoots, rooting said shoots on medium with a selective agent at a low temperature, where cold shock is used to induce callus formation in immature embryos, where low-temperature treatment of freshly picked heads of donor plants is carried out at +4 °C for 48 hours, for inducing callus formation in immature embryos, a modified nutrient medium containing 0.5 mg of 2,4-D, 2 mg of picloram, 40 g of maltose, 0.5 g of glutamine, 0.1 g of casein hydrolyzate is used, before and after transformation, the explants are kept on an osmotic medium containing 120 g/l of maltose, before transformation within 4–6 hours, and after transformation within 20–24 hours, after transformation of the explant cell, the stage of propagation (proliferation) takes place on a nutrient medium without a selective agent, rooting of shoots is carried out on a selective medium and at a lower temperature. Invention can be used to produce transgenic wheat plants with an efficiency of up to 7.8 %.
EFFECT: method for obtaining transgenic wheat plants via a bioballistics method is disclosed.
