Abstract RU 2650872 C1

FIELD: biotechnology.

SUBSTANCE: group of inventions relates to biotechnology. Claimed artificial gene encoding polyepitopic protein-immunogen MEL-TCI with properties of melanoma antigens with length of 3,428 bps. Claimed recombinant plasmid DNA pMEL-TCI which provide expression of said gene having a size of 8,845 bps. and molecular mass of 5.8×103 kDa. Claimed artificial protein-immunogen MEL-TCI containing CTL-epitopes restricted by plural allelic variants of HLA molecules of I class (HLA-A*0101, A*0201, A*0301, A*1101, A*2402, A*6801, B*0702, B*0801, B*3501, B*1801, B*4402, B*2705), and T helper determinants of melanoma antigens (NY-ESO-1, MART1, MAGE-A1, MAGE-A3, MAGE-A11, MAGE-C1), restricted by plural allelic variants of HLA molecules of II class.

EFFECT: group of inventions makes it possible to obtain artificial polyepitopic T-cellulated protein-immunogen, which, due to more optimal selection and placement of determinants provides induction of CD8 + T-lymphocytes having improved cytotoxic activity against melanoma organules.
