FIELD: molecular biology, genetic engineering.

SUBSTANCE: invention relates to the development of synthetic polyepitope vaccines against HIV-1. Artificial protein-immunogen TCI comprises CTL-epitopes as much as possible number of HIV-1 basic antigens for using as effective and safe HIV-1 DNA-vaccine. Length of artificial protein involves 392 amino acid residues (FIG. 2). The constructed protein comprises about 80 epitopes of proteins Env, Gag, Pol, Nef of HIV-1. Synthesis of artificial gene TCI (fig. 3) is carried out using polymerase chain reaction (PCR) method for amplification of HIV genome fragments encoding continuous regions of protein TCI. Genetic-engineering construction encoding the complete set of epitopes is cloned in the composite of vector expression plasmids into E. coli cells. Production of recombinant protein TCI is 10-20% of the total cellular protein level. The presence of HIV-1 proteins fragments in structure of the end immunogen is confirmed by ELISA method using HIV-1 panels of positive sera and MAT to protein p24 (fig. 2 and 3 are given in the description).

EFFECT: valuable biological and medicinal properties of gene and protein-immunogen.
