FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology and is intended to determine the index of fragmentation of sperm DNA in producer animals. Prepare a smear of sperm samples for staining and prepare the dye by mixing a solution of citric acid, sodium hydrogenphosphate and 1 % acridine orange. Dip the sample into the dye. Perform a microscopy. Spermatozoa are counted with fragmented DNA and normal, while spermatozoa with fragmented DNA are stained red, yellow or orange, normal to green. If fragmentation index of more than 30 % is detected, the sperm test is discarded and not allowed for artificial insemination of animals.
EFFECT: invention provides a simple, cheap method for determining the DNA fragmentation index.
