SUBSTANCE: relates to andrology, laboratory research methods in the field of biochemistry, pathological physiology, reproductive medicine, and can be used to clarify the causes of male infertility, as well as to predict the outcome of the IVF/ICSI procedure in assisted reproductive clinics. The ejaculate obtained by masturbation is stained. Terminal transferase is added to the ejaculate in a volume of 0.5 μl with an activity of 20-40 units/μl, the mixture is incubated in a thermostat at +37°C for 60 minutes, then add 0.1 ml of 0.05% buffered Trypan blue dye. Then, using fluorescence microscopy, the degree of DNA fragmentation of sperm is determined by counting the number of intact sperm and sperm with disturbances in the form of DNA fragmentation, calculating the ratio of damaged to undamaged sperm in %. When the degree of fragmentation is 15% or less, a low degree of DNA fragmentation is determined and male fertility is predicted with a high chance of fertilization naturally, from 16 to 30% - the average degree of DNA defragmentation is determined, male fertility is predicted with a reduced chance of fertilization naturally, more than 30% - determine a high degree of DNA defragmentation and predict male infertility.
EFFECT: improving the accuracy of infertility prognosis by determining the degree of sperm DNA fragmentation.
