CONSTRUCTION OF THE CARTRIDGE WORKING AREA FOR CARRYING OUT A POLYMERASE CHAIN REACTION Russian patent published in 2018 - IPC C12M1/00

Abstract RU 2658600 C1

FIELD: biology.

SUBSTANCE: invention relates to devices for laboratory analysis in biology and can be used in the polymerase chain reaction. Polymerase chain reaction cartridge comprises a plate of an optically transparent material with a substrate located at the bottom of the cartridge. In the body of the plate, a working zone is formed with diametrically located channels for filling the working area with reagents. Substrate is configured to heat the working area. Working area of the cartridge is in the form of a hollow cylinder with a bottom convex part bounded by a flat base facing the substrate. Convex part is formed by concentric grooves on the lateral surface, the diameters of the grooves decreasing from the periphery to the center and their dimensions lie in the range of 0.8 to 0.5 D, where D is the diameter of the cylinder and the depth along the longitudinal axis of the cylinder between the bottom and the flat base is 0.28 to 0.32 H, where H is the plate thickness. Channels for filling with reagents are made in the form of grooves and are communicated with the working area so that their bottom parts are removed from the bottom of the hollow cylinder by a distance of at least 0.3 H.

EFFECT: invention in the implementation ensures obtaining a technical result consisting in improving the quality and accuracy of the analysis by eliminating the formation of air bubbles in the working area of the cartridge both when filling with the working mixture and during thermal cycling.
