FIELD: biochemistry.
SUBSTANCE: invention relates to biochemistry, in particular to a genetically modified rodent for producing variable domains of immunoglobulin containing an amino acid histidine, encoded from its germline. Said rodent comprises in its germline an immunoglobulin locus that contains a non-arranged sequence of a variable immunoglobulin gene, comprising, in the sequence encoding the complementarity determining region 3 (CDR3), the replacement of at least one non-histidine codon with a histidine codon or insertion of at least one histidine codon. Also disclosed is a method for producing said rodent.
EFFECT: invention makes it possible to efficiently obtain domains of immunoglobulins that bind to antigens in a pH-sensitive manner.
