SUBSTANCE: invention relates to the field of biochemistry, in particular to a mouse containing in its germline a non-arranging human genomic sequence, containing a single gene segment VH person, which is a VH1-2, VH1-69, one or more gene segments DH human and one or more gene segments JH man, and inserted sequence, which encodes the mouse ADAM6a protein and the mouse ADAM6b protein, to a cell derived from it, as well as to a method for producing a rearranged human immunoglobulin heavy chain variable region sequence, method for producing an antibody and a method for preparing an antigen-binding protein of a human using the above mouse. Also a method of modifying a mouse immunoglobulin heavy chain locus is disclosed, comprising inserting one or more unrearranged human immunoglobulin gene sequences into a mouse immunoglobulin heavy chain locus, as well as modifying, which restores the activity of ADAM6 in the mouse.
EFFECT: invention makes it possible to efficiently obtain the human immunoglobulin variable region sequences.
