SUBSTANCE: present invention relates to genomic engineering. Disclosed is a method of multiplexing insertions of exogenous sequences of nucleic acids into DNA of a cell expressing RNA-directed DNA-binding protein from a type II CRISPR system, comprising introducing into a cell several hydra, complementary to different DNA sites, and several exogenous sequences of donor nucleic acids, obtaining several changes of cell DNA, and repeating said stage multiple times.
EFFECT: present invention provides introduction of multiple modifications in DNA with relatively few actions that can be further used in genome editing.
