SUBSTANCE: invention relates to cellular biology and biotechnology, namely to increase in expression of insulin and MAFA in pancreatic endocrine cells. Method includes differentiation of human pluripotent stem cells, obtained without destruction of the human embryo, or pluripotent stem cells of a person not of embryonic origin, or cells from lines of human embryonic stem cells H1 or H7, or H9, or SA002, into cells, expressing markers characteristic of the pancreatic endocrine cell line, and subsequent culturing in a medium, containing an added amount of a cyclin-dependent kinase inhibitor, which leads to an increase in the expression of insulin and MAFA.

EFFECT: invention allows to increase expression of insulin and MAFA in pancreatic endocrine cells.
