SUBSTANCE: invention refers to induction of MAFA expression in pancreatic endodermal cells, pancreatic endocrine precursor cells or pancreatic endocrine cells. Method involves culturing pancreatic endodermal cells, pancreatic endocrine precursor cells or pancreatic endocrine cells derived from human pluripotent cells of non-embryonic origin, with a medium enriched with one or more of triiodothyronine, thyroxine and thyroid hormone analogue; ALK inhibitor; or as one or more of triiodothyronine, thyroxine and thyroid hormone analogue, and ALK inhibitor, and where the medium is also enriched with one or more of Aurora kinase inhibitors, RSK inhibitors and DOT1L protein methyltransferase inhibitors.
