FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology, in particular to a process for preparing apolipoprotein A-I (Apo A-I) from a protein fraction. Process comprises suspending the protein fraction containing Apo A-I (A) having a pH in the range from 6.4 to 10.0, in a buffer solution (B) comprising 15 to 30 % of a linear or branched C1-C4 alcohol (WT/WT). Impurities are removed from the suspension while keeping the Apo A-I proteins solubilized, by precipitating by adding a linear or branched C1-C4 alcohol to a concentration of 55–65 % (WT/WT). Apo A-I is precipitated from the suspension, wherein the suspension pH is adjusted in the range from 4.6 to 5.6, and the suspension temperature is adjusted in the range from -2 to 20 °C. Apo A-I precipitate is collected.
EFFECT: proposed method is suitable for large-scale production and allows obtaining Apo A-I in high yield by suspending a much larger amount of Apo A-I without the addition of chaotropic substances.
