FIELD: medicine.
SUBSTANCE: invention relates to medicine and can be used for the manufacture of biodegradable vascular grafts of small diameter with a modified surface. For this purpose, a small-diameter hollow tubular vascular matrix is formed from the blend of biodegradable polymers based on polycaprolactone and polyhydroxybutyrate / valerate by electrospinning. Moreover, the polymers are dissolved in chloroform to a solution concentration of 5–15 %, followed by a two-stage surface modification with RGD peptides containing an aliphatic amino group. In the first step, the inner surface of the graft is treated with a 10 % solution of 4,7,10-trioxa-1,13-tridecanediamine in a 1:1 mixture of water – propane-2-ol at 37 °C for 30 min. After washing the matrix, in the second step, the inner surface of the graft is treated with a 10 mM solution of succinic acid bis-pentafluorophenyl ester in carbonate buffer pH 8.5 for 1 hour. After the second washing, the inner surface is treated with a solution of a cyclic c[RGDFK] peptide consisting of 0.2 mg/ml peptide in 50 mM carbonate buffer for 4 hours at 24 °C.
EFFECT: invention makes it possible to improve endothelization and increase the thrombore resistance of biodegradable vascular grafts.
