FIELD: biotechnology; medicine.
SUBSTANCE: invention relates to medicine, biotechnology, regenerative medicine, in particular, to methods for quantifying cells as part of a cell engineering structure (scaffold). Method for quantitative analysis of the cellular component of a scaffold is disclosed, which includes staining of samples with fluorochrome, which has a high specificity for a double-stranded DNA molecule, and fluorescence microscopy. Studied samples of scaffolds do not undergo additional preliminary sample preparation prior to staining, digital fluorescence microscopy is carried out in 10 fields of view with layer-by-layer Z-axis imaging at a predetermined depth, followed by stitching images, the obtained images are processed using filters on the intensity of fluorescent light and on the area of the object, intermediate statistical analysis is performed with the calculation of the total number of cells in each image and the average number of cells in 10 images, followed by recalculation of the number of cells in 1 mm3 of scaffold.
EFFECT: invention provides a reduction in time, labor and increase the accuracy of the study of the number of cells in the composition of the scaffold.
