METHOD FOR DETERMINING THE VIABILITY OF CELLS IN BIOMEDICAL CELL PRODUCTS IN THE PROCESS OF REGENERATION Russian patent published in 2022 - IPC G01N33/52

Abstract RU 2777257 C1

FIELD: medicine.

SUBSTANCE: invention relates to biomedicine, medicine, biotechnology, regenerative medicine; in particular, to methods for determining the viability of stem cells in biomedical cell products (BMCP) in the process of regeneration. Mesenchymal stem cells are stained with a membrane tracing dye prior to forming biomedical cell products (BMCP), the cells are included in the BMCP, the formed BMCP is cultivated in vitro, transplanted to the tissues of an experimental animal, the animal undergoes euthanasia within a control period, and tissues are isolated from the site of BMCP implantation. The tissue samples are then distributed over the bottom of a previously prepared plastic Petri dish with a diameter of 5 cm, containing 1 ml of culture medium, and visualisation and photo fixation of viable cells stained with the membrane specific tracing fluorescent dye, exhibiting characteristic fluorescent glow and morphological characteristics corresponding to mesenchymal stem cells (MSC) are performed ex-tempore, without additional sample preparation prior to visualisation. Cells exhibiting fluorescent glow and a set of morphological characteristics corresponding to mesenchymal stem cells: size from 20 to 300 mcm, presence of processes, and a visualised nucleus, are therein visualised and photofixed.

EFFECT: method reduces the labour intensity and time of evaluation of the viability of cells in biomedical cell products in the process of regeneration and eliminates errors in analysis and artifacts associated with additional impact on the cells.
