FIELD: medicine; biotechnology.

SUBSTANCE: invention relates to medicine, biotechnology, regenerative medicine, particularly to methods for assessing the direction of cell differentiation in three-dimensional scaffolds. To implement the method, the samples are stained with two fluorochromes, one of which is specific for F-actin, and the second is specific for a double-stranded DNA molecule. Cells are visualized by wide-field fluorescence microscopy, photofixation of objects with characteristic fluorescent luminescence is carried out using lenses with 10- or 20-fold magnification and an overlay function in two channels. Morphological characteristics of the detected objects are given according to the following criteria: cell shape, number of processes, degree of their branching. Conclusion on the presence of differentiation and its direction is made in the presence of cells with a certain set of signs in the analysed fields of vision.

EFFECT: method enables to characterize differentiation of cells in a three-dimensional scaffold structure.
