SUBSTANCE: invention relates to systems of multidimensional chromatography and to methods for orthogonal separation and analysis of mixtures of compounds. Typical two-dimensional reversed-phase liquid chromatography systems – supercritical fluid chromatography and methods for their use are disclosed. Claimed method and chromatography system for separating a sample contains: first separating unit comprising: a first pump unit for driving a first mobile phase through the first separating unit; a sample injector for introducing the sample into the first separating unit; and a reversed-phase liquid chromatography (RPLC) column; second separating unit comprising: a second pump unit for driving a second mobile phase through the second separating unit and a supercritical fluid chromatography (SFC) column, and a first fluid guiding unit containing a plurality of sampling loops. Moreover, the specified first fluid guiding unit is connected to the first separating block and the second separating block, where at least one of the plurality of sampling loops contains a trapping column containing a stationary phase. In this case, the chromatography system is configured to initially divide the sample in the first separating unit and subsequently inject at least a portion of the sample eluted from the RPLC column of the first separating unit into the second separating unit.
EFFECT: possibility of separation and analysis of samples by RPLC chromatography in the first dimension and by SFC chromatography in the second dimension.
