SUBSTANCE: group of inventions relates to biotechnology, specifically to the field of accurate transformation of plants, directed genome integration and protein expression in plants. Disclosed is a polynucleotide donor cassette for plant transformation, which contains a binding domain of site-specific nuclease, an analytic domain comprising a polynucleotide sequence with at least 99 % sequence identity with SEQ ID NO: 142, and a plasmid domain. Polynucleotide donor cassette is used to produce a transgenic cell for transformation of a plant.
EFFECT: inventions provide directed insertion of a universal polynucleotide donor cassette into the genome of a plant cell, fast screening of target sites and nucleases, as well as minimizing sequences that are difficult to amplify.
