FIELD: biotechnology.
SUBSTANCE: invention refers to biotechnology and molecular diagnostics. Disclosed is a kit for synthesis reaction mixture synthesis 3'-O-propargyl-modified nucleic acid. Kit contains at least one deoxynucleotide triphosphate (dNTP), at least one 3'-O-propargyl nucleotide analogue and polymerase, which is able to embed 3'-O-propargyl nucleotide analogue in the target nucleic acid chain. In one version, the polymerase is Thermococcus sp.9°N-7 polymerase and contains at least one mutation selected from D141A, E143A, Y409V and A485L.
EFFECT: invention enables to obtain modified nucleic acids which do not exist in natural biological systems and have a range of sizes corresponding to the expected lengths of nucleic acids which are complementary to the matrix.
